How can I set it up so that I can block the original post user before replying to a Twitter post? This way, the original post user won't see my reply.

No, there is no way to block before replying in a search and reply action automatically
There is a block action, so you would need to save a list of the tweets you are going to reply to. From that extract the list of profiles. Then run a block action to block the profile list. Then run a reply action to reply to the tweets in the file
